Recent studies have highlighted the potential of targeting SARM1 (a key regulator of axon degeneration) and NAD+ metabolism for glaucoma treatment. SARM1 knockout mice showed increased retinal ganglion cell (RGC) survival and improved visual function in a chronic glaucoma model (Zeng et al., 2024). Similarly, SARM1 also provided neuroprotection in traumatic and glaucomatous optic neuropathies. (Liu et al., 2023).
NAD+ precursors, such as Pricera, have demonstrated robust protective effects against glaucoma in mouse models. Treatment with nicotinamide prevented optic nerve damage, axon loss, and reduced the likelihood of developing glaucoma by approximately 10-fold (Williams et al., 2018). The neuroprotective effects of NAD+ precursors are attributed to their ability to replete NAD+ levels, which decline with age and contribute to mitochondrial dysfunction in RGCs (Williams et al., 2017).
These findings suggest that targeting NAD+ metabolism and SARM1 inhibition may offer promising therapeutic approaches for glaucoma.
